Zjawisko to jest szczególnie dobrze udokumentowane w populacjach ryb słodkowodnych zamieszkujących jeziora polodowcowe, które powstały pod koniec ostatniej epoki lodowcowej. Jednak wiedza na temat tego, jak taka zróżnicowana eksploatacja zasobów w różnych siedliskach może znaleźć odzwierciedlenie w składzie biochemicznym rozbieżnych populacji, jest nadal ograniczona, chociaż można się spodziewać takich wzorców.
W tym miejscu chcieliśmy ocenić, w jaki sposób kwasy tłuszczowe (FA) – ważny składnik biochemiczny tkanek zwierzęcych – rozdzieliły się w polimorficznym kompleksie siei (Coregonus lavaretus) i ich blisko spokrewnionych monomorficznych specjalistycznych sielawek (Coregonus albula) zasiedlających serię sześciu subarktyczne jeziora w północnej Fennoskandii. Zbadaliśmy również wzorce składu FA u drapieżników i ofiar bezkręgowców siei, aby ocenić, jak rozbieżność w ekologii troficznej między morfami siei odnosi się do profili biochemicznych ich kluczowych współpracowników sieci pokarmowej. Na koniec oceniliśmy, w jaki sposób informacje na temat dywergencji troficznej zapewniane przez zróżnicowany skład FA w porównaniu z dowodami na polimorfizm zasobów uzyskanymi z bardziej klasycznej zawartości żołądka i stabilnych informacji izotopowych (δ13C, δ15N).
Badanie zawartości żołądka dostarczyło informacji o wysokiej rozdzielczości na temat niedawno skonsumowanej ofiary, podczas gdy stabilne izotopy wskazały na szerokie wzorce zróżnicowania wykorzystania zasobów bentosowo-pelagicznych na różnych poziomach troficznych. Liniowa analiza dyskryminacyjna oparta na składzie FA okazała się znacznie skuteczniejsza w identyfikacji morfów siei i ich sielawy jako odrębnych grup w porównaniu z pozostałymi dwiema metodami. Trzy główne FA (kwas mirystynowy, kwas stearynowy i kwas eikozadienowy) okazały się szczególnie pouczające, zarówno w określaniu grup rdzeniogonidów, jak i identyfikowaniu wzorców żerowania pelagiczno-bentosowego w szerszej sieci pokarmowej.
Zawartość kwasu mirystynowego (14: 0) i stosunki δ13C w tkance mięśniowej były dodatnio skorelowane między taksonami ryb i razem zapewniały najczystszą segregację ryb wykorzystujących kontrastujące nisze pelagiczne i bentosowe. Ogólnie rzecz biorąc, nasze odkrycia podkreślają potencjał analizy FA w identyfikowaniu polimorfizmu zasobów w populacjach zwierząt, w których występuje to zjawisko, i sugerują, że ta technika może zapewnić większą rozdzielczość niż bardziej tradycyjne metody zwykle stosowane w tym celu.
Zestawy narzędzi do biologii syntetycznej do inżynierii metabolicznej sinic.
Sinice mają ogromne znaczenie dla ekologii Ziemi. Ze względu na ich zdolność do fotosyntezy i metabolizmu C1 stanowią idealne podłoże dla drobnoustrojów, które można zaprojektować do bezpośredniej konwersji dwutlenku węgla i energii słonecznej w biopaliwa i substancje biochemiczne. W celu ułatwienia wyjaśnienia podstaw biologii mikroorganizmów fotoautotroficznych i szybkiego postępu w biologii syntetycznej, opracowano zestawy narzędzi genetycznych, które umożliwiają wdrażanie nienaturalnych funkcji w zmodyfikowanych cyjanobakteriach.
W związku z tym cyjanobakterie szybko stają się wyłaniającą się platformą w biologii syntetycznej i inżynierii metabolicznej. Jednak wiele nowych typów bakterii, które zostały początkowo wzbogacone, następnie uniknęło izolacji. W związku z tym przyszłe prace hodowlane będą musiały skupić się na ulepszonych technikach subhodowli, oczyszczania i konserwacji, aby odzyskać i wykorzystać większą część różnorodności drobnoustrojów.
Aktualne wyzwania eko-metabolomiki roślin.
Stosunkowo nową dyscypliną badawczą eko-metabolomiki jest zastosowanie technik metabolomiki w ekologii w celu scharakteryzowania biochemicznych interakcji organizmów w różnych skalach przestrzennych i czasowych. Metabolomika to nieukierunkowane podejście biochemiczne do pomiaru wielu tysięcy metabolitów różnych gatunków, w tym roślin i zwierząt. Zmiany stężeń metabolitów mogą dostarczyć mechanistycznych dowodów na procesy biochemiczne, które są istotne w skali ekologicznej. Obejmują one fizjologiczne, fenotypowe i morfologiczne reakcje roślin i zbiorowisk na zmiany środowiskowe, a także interakcje z innymi organizmami.

Description: Apoptosis-Inducing Factor 1, Mitochondrial (AIFM1) is a flavoprotein essential for nuclear disassembly in apoptotic cells that is found in the mitochondrial intermembrane space in healthy cells. During apoptosis, it is translocated from the mitochondria to the nucleus to function as a proapoptotic factor in a caspase-independent pathway, while in normal mitochondria, it functions as an antiapoptotic factor via its oxidoreductase activity. The soluble form (AIFsol) found in the nucleus induces parthanatos i.e., caspase-independent fragmentation of chromosomal DNA. AIFM1 interacts with EIF3G, and thereby inhibits the EIF3 machinery and protein synthesis, and activates casapse-7 to amplify apoptosis. It binds to DNA in a sequence-independent manner and plays a critical role in caspase-independent, pyknotic cell death in hydrogen peroxide-exposed cells. |
